Test for mucopolysaccharidoses: simple method for quantitative estimation of urinary glycosaminoglycans.
نویسنده
چکیده
A simple method is described for measurement of urinary glycosaminoglycans (GAG) in normal and pathological urine: a fixed fraction of the 24-h urine is passed over a small plug of ECTEOLA-modified cellulose and the GAG are eluted with a few milliliters of NaCI solution (3 mol/liter) and measured as hexuronic acid. Because results are compared for equal aliquots of complete 24-h urines, GAG are quickly detected, thus providing means for a chemical diagnosis of mucopolysaccharidosis. The procedure requires less than 1 h, and several tests may be conveniently run simultaneously. When urine specimens collected from normal children or children affected by mucopolysaccharidosis were analyzed, the results obtained with the proposed procedure agree well with those of more elaborate, widely accepted methods.

1. The effect of MgCl(2) concentration on the interaction of Alcian Blue 8GX and glycosaminoglycans in the urine of patients with mucopolysaccharidosis was studied by using a new quantitative micro method for the measurement of Alcian Blue-glycosaminoglycan complexes. This provided a means of measuring the critical electrolyte concentrations of urinary glycosaminoglycans. 2. Theoretical conside...
متن کاملMeasuring urinary glycosaminoglycans in the presence of protein: an improved screening procedure for mucopolysaccharidoses based on dimethylmethylene blue.
Earlier we described a simple and reliable screening procedure in urine for mucopolysaccharidoses based on the color reaction of glycosaminoglycans (GAGs) with dimethylmethylene blue. At physiological concentrations of urinary protein, we observed an obvious interference by protein in the assay. By modifying the assay, we abolished the protein interference. The modified procedure is not disturb...

A considerable need exists for improved biomarkers for differential diagnosis, prognosis and monitoring of therapeutic interventions for mucopolysaccharidoses (MPS), inherited metabolic disorders that involve lysosomal storage of glycosaminoglycans. Here we report a simple, reliable method based on the detection of abundant nonreducing ends of the glycosaminoglycans that accumulate in cells, bl...
متن کاملAnalysis of glycosaminoglycans in urine by using acridine orange fluorescence.
The fluorescence technique described here utilizes the electrostatic interaction between the polyanionic sites of glycosaminoglycans and the cationic dye Acridine Orange to analyse urinary glycosaminoglycans from patients suffering from mucopolysaccharidoses. The basis of the titration is the decrease in the fluorescence of free Acridine Orange that occurs when it is bound to polyanions. The ef...
متن کاملQuantitative estimation of lysosomal storage in mucopolysaccharidoses by electron microscopy analysis.
Mucopolysaccharidoses (MPS) are severe inherited metabolic disorders caused by storage of glycosaminoglycans (GAGs). The level of accumulated GAGs is an important parameter in assessment of the severity of the disease and the efficacy of treatment; unfortunately, biochemical methods are often unreliable and only semi-quantitative. Therefore, finding other methods for estimation of GAG levels an...
